Part 1: Obervation of neural precursor cells in rat dorsal root ganglia, superior cervical ganglia and trigeminal ganglia.AIM:To investigate neural precursor cells in rat dorsal root ganglia(DRG),superior cervical ganglia(SCG),trigeminal ganglia(TrG). METHODS:the ganglia from embryonic and adult rats DRG, TrG and adult DRG,TrG,SCG were sliced for Brdu,Nestin,P75 immunofluorescence histochemical test.Additionally, the ganglia were respectively cultured and observed in their proliferation and differentiation. The cells were identified by Brdu,Nestin,P75,NF and GFAP immunofluorescence histochemical test.RESULTS:In the frozen sections, Brdu/Nestin and Nestin/P75 positive cells were found. The cultured cells could divide and proliferate and showed Nestin, Brdu,P75 positive .The third-passage cells could differentiate in the presence of fetal bovine serum.The differentiating cells showed NF and GFAP positive .CONCLUSION:There are neural precursor cells that can divide and proliferate in embryonic, neonatal rat DRG. TrG and in adult DRG.,TrG and SCG They can differentiate into neuron and glia.cells.Part 2: Effect of pain stimulation on neural precursor cells'proliferation and differentiation in adult rat dorsal root ganglia.AIM:To evaluate if pain stimulation can inspire neural precursor cells'proliferation and differ- entiation in adult rat ganglia. METHODS:1% formalin were injected into model's left forelimb.12 rats grouped into 3 teams (4 rats in every team ):control team,3 days team of pain,7 days team of pain.All teams were continuously marked with Brdu.Then they were killed at different time point.DRG of contral team and 3 days team were done Brdu/Nestin immunofluorescence test to compare the Brdu/Nestin cell number in pain pathway with nopain pathway and the number of contral team. .DRG of 7days team were done BrdU/NF and BrdU/GFAP immunofluorescence test to compare the double-labelled cell number in pain pathway with onpain pathway. RESULTS: Brdu/ Nestin positive cells could seen in DRG of contral team and 3 days team.There was no obvious difference in the double-labelled cell number between pain pathway and nopain pathway.In 7 days team, BrdU/GFAP positive cells were obvious more than BrdU/NF positive cells. CONCLUSION:Efffect of pain stimuliation on neural precursor cells'proliferation in adult rat DRG is not very distinct.Those proliferating precursor cells in adult rat DRG will differentiate into glia cells.
