Effects Of Puerarin And/or Fluvastatin Pretreatment On Acute Myocardial Ischemia/Reperfusion Injury Of Rabbits

Objective: To investigate whether short-term Puerarin and/or Fluvastatin pretreatment can protect myocardium suffered from rabbit acute myocardial ischemia/reperfusion injury (IRI). Methods: Sixty rabbits were divided to 5 groups randomly(n=12): The Sham operated group (S group) were administered with capsula cordis clipping and braiding without ligatating conorary and medicine pretreatment. The Control group (C group) were suffered IRI without medicine pretreatment. The Puerarin group (P group) was pretreated with intravenous injection of Puerarin in dose of 20mg/kg/d for 3 days and 20mg/kg before operation. The Fluvastatin group (F gruop) was pretreated with oral administration of Fluvastatin in dose of 20mg/kg/d for 3 days and 20mg/kg before operation. The Puerarin + Fluvastatin group (P+F group) was pretreated with intravenous injection of Puerarin in the same dose of Puerarin group and oral administration of Fluvastatin in dose of 20mg/kg/d for 3 days and 20mg/kg before operation. After two hours reperfusion ,drawing 4.5ml blood of atrio dextro(AD) from jugular, adding the blood into the aprotinin tubes of 10%EDTA, centrifugalization in 3000rpm and 4℃ for 10 minutes, separting the plasma, and conservation the plasma in -70℃.then killing the rabit, taking out of the heart, microtome section, taking cardic muscle slices into TTC for 30 minutes, immobility in 10% formalin for 24 hours, then take a photo, analysis by photoshop8.0, evaluation...
