Pentachloronitrobenzene (PCNB) was explored by Olin Mathieson chemical company in the recently fifty decades, and its common name was quintozene. It was a kind of organochlorine pesticide, belonging to phenylated fungicides. PCNB had many chemical properties, such as stable chemical property, low vaporability, uneasy to be break down, long residual period, not to be dissolved in water, and was easily dissolved in organic solvent and fat. And if PCNB was taken into body, it could be accumulated in the tissues and organs which are high in fat content, and caused chronic poisoning and all kinds of specific diseases. PCNB had been applied in the preventions of vegetable's germina and seeds diseases and insect pests as a sort of soil disinfectants. In the growing process of some effective plants such as ginseng and ginkgo, PCNB was used a large amount of application, and led to a high residual dose in those health promotion foods and products quality reduction, so it had become one of the production quality test indicators of ginseng and aralia quinquefolia.Some studies indicated that main harm of PCNB was liver, kidney and neural system damages, and its influence of genital system hadn't been reported yet. Along with many types of pesticide's large amounts of application, environmental and diet pollutions were pricked up continually. As an important component of the body-genital system was also a target organ that is easily be attacked by exotic chemicals. This study was aimed to discuss the effects of PNCB toxicity on reproductive system of male mouse and its potential mechanisms. The study was wanted to discuss the effects of PCNB toxicity on reproductive system of mouse through six aspects of influence, that were organ coefficient, rate of sperm deformation, testicle lipid peroxidation, special enzyme, pathological changes, and apoptosis, in order to provide basis for reproductive function harm of male and mammal reproductive toxicity of PCNB.Methods: Totally 60 mice were divided into five groups (natural control group, solvent (peanut oil) control group, and low dose group (100 mg/kg), middle dose group (300 mg/kg), high dose group (500 mg/kg)) followed the random principle, and used intragastric administration once a day lasting for 21 days. After those days, those mice were killed and taken liver, kidney and testicle, to detected organ coefficient of the above organs, spermatozoon abnormal form quotient, quantity changes of H2O2, MDA in testicle, quality variety of SOD, LDH, GSH, GSH-Px ,G-6PDand GST, pathological changes, and apoptosis index of mouse testicle.Results: (1)Influences of mouse organ coefficient: In this study, organ coefficient of liver was risen along with the poisoning doses, and had remarkable difference between control and middle, high groups. And there were congestion, hydroncus and hyperplasia in the liver. But organ coefficient of kidney and testicle didn't have the trendency, having no remarkable difference between control and other groups. (2)Impacts of rate of sperm deformation: Along with the poisoning increased, rate of sperm deformation was risen, and had remarkable difference between dose and control groups.(3)Affects of testicle lipid peroxidation: Quantities of H2O2, MDA were increased and qualities of SOD, LDH, GSH, GSH-Px, and GST were decreased obviously with the risen poisoning dose.(4)Influences of special enzyme: In this study, along with the increased dose, quality of LDH, G-6PDH was reduced gradually.(5)Impacts of pathological changes: Structure of convoluted tubule was changed obviously-cells arrangement loosened and clearance largened, with the increased dose(.6)Affects of sperm cells apoptosis: TUNEL method was used to detected apoptosis of spermatogenic cells, and the study indicated that apoptosis index increased, but the apoptosis rate decreased when injury exacerbated.Conclusions: (1) PCNB had reproductive toxicity to mouse, and dose-reaction relation, could increase rate of sperm deformation and testicle cell apoptosis index, pathological changes, resulted in decreasing of germ cell quantity. (2)PCNB could cause pathological changes of multi-organs. (3) PCNB had oxidation damage to mouse testicle. (4) Mouse testicle was one of the target organs of PCNB.
