The brown alga Ishige okamurae and the green alga Ulva conglobata are large-scale economic algae of South China sea. The chemical constituents of Ishige okamurae and Ulva conglobata, sulfated polysaccharides from Ishige okamurae and their bioactivities were investigated.Three compounds were isolated from Ishige okamurae, in combination with the bioassy-guided method. Their structures were detected by chemical and spectral method. Two compounds were identified as hexadecanoic acid (I), Saringosterol(II),fucosterol (III). One compound from Ulva conglobata was obtained and identified as hexadecanoic acid. Octadecanamide, hexadecanamide and 1,2 benzenedicarboxylic acid diisooctyl ester was detected by the GC-MS.The anti-virus activities of extracts and fractions from Ishige okamurae and Ulva conglobata against HSV-1,Cox B3 in vitro were studied by MTT and CPE method. As a result, the most of the extracts and fractions showed obvious cytotoxicities. The cytotoxicity of chemical extracts and fractions and one compoud from Ishige okamurae and Ulva conglobata to H460 cell was tested by CPE method. The results indicated that the CC50 of the both acetic acid ester fractions of Ishige okamurae and Ulva conglobata is 31.25μg/mL, the CC50 of the compoud also is 31.25μg/mL.Sulfated polysaccharides of Ishige okamurae were extracted with hot water, separated by DEAE column chromatography. The functional groups, the amounts of monosaccharides and sulfate groups of the polysaccharides were determined. As a result, the fucose is the main monosaccharide of the crude polysaccharides, covering 67.05%. The relative contents of sulfate groups is 10.30%.
