| Object: To construct a simple model of n-hexane inhalation in rats, and provide an experimental model of lung, liver and kidney damage. To observe the expressions of relative factors in lungs after sub-acute n-hexane inhalation, and study the mechanisms of lung damage during the course.Methods: 24 healthy male rats were selected and randomly divided into 4 groups: control group, n-hexane groups (l,3and 7 days). Primary concentration of n-hexane was 35.2g/m~3, 8 hours per day. The lungs, livers and kidneys were taken out soon after the rats were killed, then were embedded by paraffin, sliced and stained by haematoxylin-eosin in the routine way, and the slices were read by microscope. And took the immunohistochemistry researches of lysozyme, surfactant protein B, neuronspecific enolase ,nestin and calcitonin gene related peptides.Result:The average blood n-hexane concentration of each n-hexane group (l,3and 7 days) were 636.92ug/L,913.49ug/L and 1090.69ug/L.There were primary pathologic damage of lungs, kidneys and livers in rats after n-hexane inhalation.In the rats lungs of n-hexane groups, the respiratory tract epithelium cells were found damaged and partly ablated. Meanwhile the positive cell numbers of Lysozyme increased along with the time of n-hexane inhalation; and the positive cell numbers of surfactant protein B increased within 3 days, and reduced in 7-day groups; The positive cells of neuronspecific enolase and nestin scattered in the epithelium of respiratory tract, and the positive cell numbers increased in 7-day group than control group. The positive cell numbers of calcitonin gene related peptides increased along with the time of n-hexane inhalation, and the coloration of positive matters in adventitial coat of arteries and veins were stronger in 7-day group than the control group. |
