Objectives:Study the method of amniochorionic membranes cultured in vitro,the mechanism that infection cause preterm labour ,and the ability that N-acetylcysteine(NAC) prevent preterm labour caused by infection .Methods:Amniochorionic membranes were collected from 20 pregnancy women undergoing elective cesarean delivery at term,without evidence of active labour and complications of pregnancy,then these amniochorionic membranes were cultured in vitro for 72 hours .When these amniochorionic membranes were cultured for 48 hours ,some were treated with LPS( Lipopolysaccharide )or LPS+ NAC .All these cultured amniochorionic membranes were divided into 5 groups :0 hour group;24 hour group;48 hour group;72 hour LPS group;72 hour LPS+NAC group.At the end of cultures, the viability of these cultured membranes were assessed ,and then determined the expression of MMP-9 and TIMP-1 in all groups by Immunohistochemical and RT-PCR ,finally the ratio of MMP-9/TIMP-1 in LPS group and LPS+NAC group was compared .Results:The viabilitiy of cultured amniochorionic membranes in 24h group,48h group,72h LPS group and 72h LPS+NAC group was 87%±1.9%,85%±1.3%,83%±1.6%,82%±1.2%,whether Immunohistochemical or Reverse Transcription Polymerase Chain Reaction (RT-PCR) ,we can hardly see the expression of MMP-9 in 0 hour group and 48 hour group ,the expression of MMP-9 in 24 hour group was slightly elevated , it was strongest in 72 hour LPS group , the expression of MMP-9 in 72 hour...
