Background: As a part of panaxadiol saponin, ginsenoside has many functions such as central inhibition, anti-low temperature, degrading the concentration of intracellular Ca2+,antiocxygetation and anti-aging.At present ,it is has been proved that Rb1 can constitute calcium channel in myocardial cell, degrade the concentration of intracellular Ca2+ and protect the animal pattern with myocardial ischemia and myocardial failure.However whether Rb1 can antagonize hypertension and reversal the change of a serial of myocardiac mechanics ,nerves and humor in the development of hypertension.Objective: our research is the first to demonstrate that whether ginsenoside Rb1 can antagonize hypertension and block the formation of hypertension , also to investigate how Rb1 protect the cardiac in the hypertension. We get dose effect curve when we study the Rb1 effect on the acute and chronic hypertension and it gives some theory evidence about clinical explanation.Method: we use NE and LNNA to construct the rat hypertension model and then study the function that Rb1 of different dose have on the arteriotony ,myocardiac mechanics and blood biochemical indicator of the hypertension model .Result :1.Defferent dose all can antagonize acute hypertension caused by 0.01%NE and resulted in the depression of heart rate and the blood pressure and lowered myocardiac contractility thus smooth over the adrenergic never adverse reaction. The depressurization effective of Rb1 has dose dependent and with the dose added, the effective on the hypertension is more notble. At 30mg/kg there is the best effective and induced the heart rate to be reduced by12.8% , systolic pressure and diastolic pressure by 12.5% and 16.3% .At the same time the cardiac max contractility and relaxtion have also been reduced by 12.5% and 16.3% .2 Rb1 can reduce chronic hypertension caused by LNNA with dose-dependent and protect the hypertension cardiac diastolic function disorder. At 30mg/kg there is the best effective and resulted in systolic arterial pressure and diastolic pressure ofthe model reduced by 17.1% and 18.3%. LVSP and LVEDP also reduced by18.1% and 22.9%,at the some time the max contractive rate and diastole rate reduced by 37.8% and 41.2%,return the normal level.3 Rb1 also can suppress the lipid peroxidation activation that caused by the augment of oxygen free radical because of hypertension, and it can raise SOD and reduce the MDA in the blood ,reduce CK then can protect the damage because of hypertension .This kind of protection also has dose-dependent ,and at30mg/kg, Can raise SOD by 23% , reduce MDA by 47%and CK by 26.3%,it is indicate that is the most effective dose.
