Fuling, the sclerotium of Poria cocos, is one of the most important traditional herbs in China. It mainly contains polysaccharide, terpene, ergosterol, protein and so on. In traditional decoction method or modern pharmacy industry for Chinese medicine, the effectual components in Chinese herbs are extracted with water or alcohol generally. Pachyman, which takes up about 84% of Fuling net dry weight, is usually discarded after decoction and extraction, because of its water and alcohol insolubility.In order to take good use of pachyman and to increase the utilization ratio of Fuling, some pachyman ramifications, such as carboxymethyl pachyman and sulfated pachyman, which have the antitumor and immunity enhancing bio-activities, are obtained by chemical modification. However, during the chemical modification process, there are some disadvantages, such as harmful leftovers to environment and so on. In this subject, pachyman is hydrolyzed with β-glucanase to get water soluble pachyman(WSP), and the physical and chemical properties and antitumor activities of WSP are studied. The content concludes three main parts as following:I . Studies on the hydrolyzation conditions of pachyman by enzymes. A steady pachyman suspension was prepared by NaOH-HAc pre-operation. And No.C enzyme was selected from commercial enzymes for hydrolyzation. The optimal hydrolyzation conditions of No.C enzyme were 55℃, pH 4, substrate concentration 4%, enzyme added 2U/mL and hydrolyzation time 6h, in which the rate of WSP was up to 57.9%.II. Studies on WSP purification and its physical and chemical properties. WSP was divided into two major components, WSP-1 and WSP-2, through SephadexG-100 column.
