Object To found a different cDNA library with Malasesiza furfur in hypha phase, and procure its gene fragment on differential expression. Method First, to isolate the total RNA both in hypha and yeast phase of Malasesiza furfur, Then reverse them to cDNA and found a different cDNA library with Malasesiza furfur in hypha phase by Subtractive Hybridization (SH); At last, clone and sequencing the gene fragment on differential expression of Malasesiza furfur in hypha phase.Result 1.Gain the RNA which can fit many molecular biology technologies; 2.Found a different cDNA library with Malasesiza furfur in hypha phase; 3.Gain many clone of gene fragment on differential expression, choice 3 of them for sequencing, and blast the sequence in Genebank. Conclusion Found a different cDNA library with Malasesiza furfur in hypha phase by Subtractive Hybridization (SH); And sequencing some fragment of differential expression gene. Provid foundation for the further study on complete differential gene expression.
