Differentiation is very important in the growth and development of human and other multicellular organisms, it is neither always abnormal nor always detrimental. Deregulation of cell differentiation is associated with the pathogenesis of many kinds of diseases. The genesis of leukemia is usually because of the abnormality occurring in the process of the differentiation of hematopoietic stem cells and it caused the infinite proliferation of the stem cells that cannot be end-differentiation. Study of the mechanism of cell differentiation help us perceive the essential rules in life and the genesis mechanism of neoplasm. The further understanding of cell differentiation always follows the cloning of the novel gene related to differentiation. So it has profound significance to clone differentiation related genes. In this study, the chronic myeloid leukemia cell line K562 was induced to macrophage-like differentiation by Hexamethylene Bisacetamide (HMBA), this cell differentiation model was not reported very often. The genes differentially expressed in the differentiated K562 cells were cloned by using the cDNA subtractive hybridization method. We got 64 clones, there were 53...
