Objective To investigate the effects of CO2 pneumoperitoneum on pancreatic function in diabetic rabbits.Methods Forty three Japan white rabbits were randomizedly divided into two groups: four in normal control group(No), others were made diabetic model with Allxon. Thirty six were succeed in creating diabetic model and were randomizedly divided into three groups as designed for experimental groups: Rabbits in two experimental groups were subjected to CO2 pneumoperitoneum with an intraabdominal pressure of 10 or 15 mmHg for 2 hour (T10/T15) while rabbits in experimental control group without pneumoperitoneum(T0). After Femoral artery were cannulated successfully, CO2 pneumoperitoneum of different pressures were established by closed method. Artery blood samples were collected for measuring blood glouse, amylase and concentrations of insulin, C-peptid by radioimmunoassay, which were repeated before the onset of CO2 pneumoperitoneum, 0,2,6,12 hours after deflation. Then rabbits were killed. The pancreas were removed and divided into two parts: the tail of pancreas fixed in a 10% buffered formalin solution, and then observed by conventional light microscopy, the head of pancreas were prepare homogenate, which was used to detect SOD activity by Xanthineoxidase way and MDA content by thio-barbituric acid way.Results (1)After establishment of diabetic model, compared with normal control group the level of blood glouse, amylase,MDA content were increased, concentrations of insulin, C-peptid and SOD activity were decreased. (2) After abdominal deflation, there was significant increasing in blood glouse, amylase, insulin, C-peptid, MDA content and significant decreasing in SOD activity. Until 12 hours after abdominal deflation, the level of blood glouse, amylase, insulin, C-peptid, MDA content returned to formal level in group T10, but still did not return in group T15. and SOD activity...
