k-carrageenan(KC) is a kind of marine acidic oligosaccharides made ofβ-(1→3)-D- galactopyranose (Galp) -4-O-sulfo group andα-(1→4)-3,6-anhydro-D- galactopyranose (AnGalp), it has many bio-activities, as anticoagulant, anti-virus, improving of immunity, antitumor and anti-inflammation. Also k-carrageenan was applied extensively in health food. At the same time, there are some comparabilitis in structure and the properties of electrification between k-carrageenan and endogenous substances, so it have many potential activities and applied value.High performance capillary electrophoresis(HPCE) is a new analytical technology which has been developed in the last twenty years. It has been applied in many areas for its celerity, efficiency, high sensitivity ,low experimental consumption and less sampling amount. HPCE has begun to be analyze saccharides at the end of the eighties and been developed very fast. In this paper, oligosaccharides of k-carrageenan have been separated and analyzed by CZE-UV of high performance capillary electrophoresis (HPCE).Theκ-carrageenan was degraded through acid hydrolysis and purified by Bio-Gel-P6 according to their molecular weight at first. K-carrageenan oligosaccharides degraded by acid are dirivatized with 8-aminonaphthalene-1,3,6-trisulfonic acid Disodium salt(ANTS) by reductive amination using capillary zone electrophoresis by pre-column derivatization , their ANTS derivatives are separated and detected by capillary zone electrophoresis with UV. The derivatization reaction conditions, including the concentration of ANTS, pH value of derivatization buffer, reaction time, temperature and separation conditions are investigated in detail.
