The focus of this research is to study the influence of baicalein and baicalin on porcine preadipocyte proliferation, differentiation and transcription of FAS and transcription factors (PPARγ2,C/EBPα).In the research, SV cells from subcutaneous fat was adopted to separate preadipocytes, and the cell biological characteristics of preadipocytes from differente age donor was studied to choose the best donor for preadipocytes which had better states of proliferation and differentiation.On the base, the methods of morphologic observation, Oil Red O straining, MTT spectrophotometry, spectrophotometric analysis, and semiquantitative RT-PCR were applied to determine the effects of baicalein and baicalin on the proliferation and differentiation of porcine preadipocytes. At the same time, the mechanism of its effect was discussed principially. This research includes following several aspects:1 To study the proper donor that can make the porcine preadipocytes reach better state of proliferation and differentiation, SV cells from 90 day old pig fetuses, young pigs(1-3 day ) and 120 day old pigs adipose tiuess were adopted. The proliferation and differentiation of preadipocytes were determined by MTT spectrophotometry and Oil Red O staining respectively. The results showed that SV cells from 90 day old pig fetuses and 1~3 day old pig benefited for proliferation and differentiation of the preadipocytes.2 To study the effect of baicalein and baicalin on proliferation of porcine preadipocytes, MTT spectrophotometry and FCM(flow cytometry) were adopted. The Results showed that 160~640μmol/L baicalein and baicalin inhibited the proliferation of preadipocytes in a dose dependent manner(P<0.05). The concentration of baicalein at 40μmol/L had no effect on adipocyte proliferation. However, 20μmol/L baicalin promoted the proliferation of preadipocyte and the promotive effects correlated with the concentration of baicalin negatively.3 To study the effect of baicalein and baicalin on the differentiation of porcine preadipocytes, accumulation of TG(triglyceride) in the adipocytes was determined by Oil Red O staining, and the cell percentage and degree of lipid accumulation were determined by...
