In recent years,studies have shown that miRNA is involved in adipocyte differentiation and lipid metabolism.This study select the mature adipose cells 3T3-L1 cells which study on fat metabolism disorder as for the research object of this experiment,in order to detect the effects of miR-26a-5p on 3T3-L1 preadipocyte cell differentiation and proliferation.Following the transfection of miR-26a-5p mimic and miR-26a-5p inhibitor into 3T3-L1 preadipocyte cell,CCK-8 was used to detect the cell proliferation and flow cytometry to detect the cell cycle;selected cell cycle genes CyclinD1 and CDK4 and adipogenic related genes C/EBP? and FABP4 were detected their expression by qRT-PCR and Western blot;using oil red O staining to determine the accumulation of triglycerides.The following results are obtained:1.miR-26a-5p regulates 3T3-L1 preadipocyte cell proliferation: overexpress of miR-26a-5p inhibits the proliferation of 3T3-L1 preadipocyte cell,resulting in cell cycle genes CyclinD1 and CDK4 were significantly down regulated at the mRNA and protein level;while inhibition of miR-26a-5p promotes proliferation of 3T3-L1 preadipocyte cell,resulting in cell cycle genes CyclinD1 and CDK4 at the mRNA level and protein level were significantly and very significantly increased.2.The expression of miR-26a-5p during adipogenic differentiation of 3T3-L1 preadipocyte cell: the expression level of miR-26a-5p was time-dependent,which was significantly up-regulated during the differentiation of 3T3-L1 preadipocyte cell.3.miR-26a-5p regulates 3T3-L1 preadipocyte cell differentiation: overexpress of miR-26a-5p promotes 3T3-L1 preadipocyte cell triglyceride accumulation,C/EBP? and FABP4 level of mRNA and protein was significantly or very significantly increased;the inhibition of miR-26a-5p showed the opposite situation.Conclusion: miR-26a-5p inhibits 3T3-L1 preadipocyte proliferation and promotes adipogenic differentiation.
