Objective To explore the effects of losartan on renal inflammatory injury and podocyte injury of the rats with diabetesMethods Divide the Wistar rats into three groups: normal group(NC),diabetes model group(DM) and diabetes group with Losartan treatment(DL).⑴Observe body weight, kidney weight, kidney/body weight ratio , 24h urinary protein excretory ratio,serum creatinine(Scr) ,BUN and the changes of the pathologic feature of the kidney.⑵By immunohistochemistry staining, observe the expression of the ICAM-1,CD45, and VEGF of kidney tissue section.⑶By wetern blotting ,observe the expresson of nephrin(one marker protein of podocytes) and VEGF of the kidney. tissue.Results Losartan inhibits the significantly increaced expression of ICAM-1 ,CD45 and VEGF in renal tissue of rats with diabetes and relieves the significantly decreased expression of nephrin, as well as reduces the significantly increased level of the kidney weight , kidney weight/body weight ratio, 24h urinary protein excretory ratio and attenuates the renal pathologic lesions such as podocyte foot process fuse and GBM thickening. Those pathlogic and clinic changes are also observed in the diabetes rats without Losartan treatment ,but have changed more seriously.
