Objective: To explore the role of MMPs/TIMPs alterations in the pathogenesis of diabetic cardiomyopathy in type 2 diabetes(T2DM) rats.Methods:20 male Wistar rats were randomly divided into normal control group and T2DM group, each had 10 rats. T2DM rats were induced by small dose injection of streptozotocin and being fed with high-fat-diet. Euglycemic-hyperinsulinism clamp technique (EICT) to test the insulin resistance was performed and insulin induced glucose infusion rate(GIR) was used to evaluate the insulin sensitivity.The body weight(BW),heart weight(HW) and HW/BW were measured. The ultrastructural changes were evaluated by electron microscope and myocardial apoptosis was detected by TUNEL.The mRNA expression of MMP-2, MMP-9, TIMP-1, TIMP-2 were determined by RT-PCR. The protein level of MMP-2, MMP-9, TIMP-1, TIMP-2, collagenâ… a nd collagenâ…¢were detected by immunohistochemistry.Results:In the T2DM group, GIR significantly decreased, while body weight increased as compared with the normal control group(p<0.01).
