Objective:1. To provide reasonable and practicable animal models for clinical cerebral ischemia research.2. To investigate the area of infarct, the water content, SOD activity, MDA content and the expressions of NF- κ B, I κ B α , MMP-9 in rats following focal cerebral ischemia-reperfusion.3. To explore the protective effect of grape seed procyanidinds extract and the effect on SOD activity, MDA content and the expressions of NF- κ B, I κ B α , MMP-9 in rats after focal cerebral ischemia-reperfusion.4. To provide theory and experiment evidence for brain protective therapy following cerebral ischemia-reperfusion.Methods: In this paper, the models of focal cerebral ischemia reperfusion in rats were established suture-occluded method. The area of infarct was examined by 2,3,5-triphenyltetrazolium chloride (TTC) staining. The water content was calculated by dry and wet weight method. Superoxide dismutase (SOD) activity and malondialdehyde (MDA) content were measured. The expression of NF- κ B, I κ B α and MMP-9 were examined by immunohistochemical method (SABC).Results: (1)Compared with focal cerebral ischemia-reperfusion group, the area of infarct in (100mg/kg and 200mg/kg) GSPE (Grape seed procyanidins extract) therapeutic groups decreased significantly at 24h after reperfusion (P<0.01) .
