To elucidate the underlying mechanism of allergic reaction to penicillins and explore the relationship between cross-reactivity and chemical structure of the penicillins, we used radioallergosorbent test (RAST) to examine 8 kinds of specific IgE antibodies of different antigenic determinants in the serum of 195 patients with allergic reaction to penicillins. RAST inhibition studies were carried out to identify the fine structural features recognized by IgE antibodies. Freshly isolated peripheral blood mononuclear cells of penicillin-allergic patients were stimulated with penicillins in order to understand the molecular mechanisms of cross-recognition of various penicillins by peripheral blood lymphocyte cells. 1. Associations between specific IgE antibodies and skin test1.1 Associations between the positive rate of specific IgE antibodies and skin testWe grouped 195 patients into group with positive skin test and that with negative skin test. The positive rates of IgE antibodies among 138 patients with positive skin test and 57 patients with negative skin test were 66.67% and 59.65%, respectively.1.2 Associations between kinds of specific IgE antibodies and skin testRAST was used to examine 8 kinds of specific IgE antibodies to major and minor antigenic determinants of 4 drugs (PG, PV, AMP, AX).The positive rate of IgE antibodies to PG among 138 patients with positive skin test was significantly higher than that to PV (P<0.05), but there was no significant difference between 4 drugs (P>0.05).
