Rheumatoid arthritis(RA) is an autoimmune disease characterized by progressive joint destrution. The pathology change of RA is synovitis, destructions of cartilage and bones. The important mechanism of RA is lymphocyte's abnormal activation and homing to synovium. It has been proved the lymphocyte's infiltration to articular cavity has close relations with destructions of cartilage and bones. Suppression of lymphocyte's inflammation homing is the important element of effective treatment.Human leukocyte antigen-G (HLA-G) belongs to nonclassical HLA-I. It expresses at trophoblast of mother-fetus interface. It participates immune tolerance.In normal, that is important factor which does not cause mother'rejection to fetus. It is necessary to the success of encyesis. Recently, researches have proved HLA-G has relations with organ transplantations, reproductions, tumors, etc. It is an important agent which causes immune tolerance. It can suppress lymphocyte CD4+T cells directly, induce apoptosis of CD8+T cells, suppress the cytolysis of NK cells, which can influence the immunity reaction.Objective Study the effect of peripheral-blood lymphocyte inhibition induced by the recombination of HLA-G in patients with rheumatoid arthritis, through constructing recombination HLA-G eukaryotic expression plasmid, transfect NIH3T3 mouse collagenoblast cell line.
