Cartilage injury is one of the reasons which induce osteoarthritis in the end. Healing of such kind of injury can reduce the rate of OA. Traditional treatment has much shortcoming. Tissue engineering is a promise technology, which may solve the problem of cartilage repair. Although the future of this technology is bright, there are still many problems needed to be resolved. Loss of cell phenotype during the culture of seed cells in vitro is one of the difficulties, which limits the development of cartilage tissue engineering research in some degree.Objectives Relying on the technology of environment simulation and environment optimization, the best environment was investigated in which seed cells can express its phenotype stablely. The experiment includes three parts: environment analyses; environment simulation; environment optimization, and pays more attention on the effect of cell cytokine and serum concentration. Simulation internal environment of articular by environment simulator which was manufactured by myself. According to well-distributed analyses, the optimization environment was sieved.
