Clinical Study Of Weekly Dose Of Irinotecan Combined With Low Dose Of FP Regiment In The Treatment Of Patients With Metastatic Colorectal Cancer

Objective:To evaluate the efficacy , the survival duration,the clinical benefit response and the adverse effects of weekly dose of combination chemotherapy of Irinotecan (CPT-11) and low dose of FP regiment for the treatment of patients with metastatic colorectal cancer.Methods :21 patients with metastatic colorectal cancer were treated by IFP regiment. CPT-11 60 mg/m~2 for 30 minutes on days1 ,8and 15 were administered by intravenous infusion in conjunction with low dose of 5-FU 300mg/m~2/d,by continuous infusion for 14 days and low dose of cisplatin 5 mg/d,for 5 days every week until 2 weeks. The course above was repeated every 4 weeks. The efficacy was evaluated after 2 cycles.Results: Among 21 patients with advanced colorectal cancer were all retreatment, 20 patients were eligible for response evaluation underwent chemotherapy. There were 4 (20.0%) objective response, because 4 partial responses. The overall response rate was 20.0%, the median time to progression was 5 months. the median survival time was 15 months. The rate to benefit was 75.0% .21 patients were eligible for toxicity evaluation underwent chemotherapy .The main side effect included acute cholinergic syndrome , late-onset diarrhea and neutropenia . Acute cholinergic syndromemainly espressed mild that observed in 4 patients (19.0%), neutropenia mainly expressed grade I to II ,the ratios of neutropenia was 42.9%.
