Senile dementia (SD) has been a major threat to the health of human beings over a long period of time. However, the etiopathogenisis and pathogenesis of this disease as well as its therapeutical approach still remain poorly understood. Goutengsan is a traditional Chinese prescription from "Benshi Fang", mainly treat liver-heat and syncope due to disorder of the liver,dizziness and blurred vision,distending pain in the chest and diaphragm,restlessness and fever with thirst, etc. Recently, several lines of reports had shown that Goutengsan was effective against SD in clinical practices, however, its exact therapeutical value and mechanism were not completely investigated. In present study, an experimental animal model of Alzheimer dementia(AD) was employed to assess the therapeutical effect of Goutengsan on AD and its possible mechanism.1. safety evaluation:The LD50 of Goutengsan extract was hard to be determined for its extreme low toxic property, thus its maximal tolerance dose was measured. The experimental procedures were described as follows: 20 mice after fast for 16 days were intra-gastric administrated with Goutengsan extract (2.625g/ml) at maximal dosage of 0.5ml/10g. No death was found in 7 days after administration. Then another 30 mice were single-dose administrated with maximal dosage of Goutengsan extract to evaluate its acute toxic reactions. The results demonstrated that no significant toxic signs were observed after drug administration. The maximal dosage in mice was equivalent to 262.5 times the dosage used in human beings, thus ,this extract was safe enough to be used in clinical practices.2. wisdom promoting effect of Goutengsan extract120 mice were randomly divided into 6 groups, 3 experimental groups of which were daily administrated (i.g.) with Goutengsan extract at high, middle and low dosages respectively. The model and control groups were administrated with D.D.W whereas aniracetam at 0.2 g/kg body weight served as a positive control.
