In recent years,Alzheimer's disease(AD)has increased year by year,so finding a biomarker to conduct early biological diagnosis of AD is a current research focus.Based on the 3D-ASL sequence,this paper explored the role of Cerebral blood flow(CBF)in the pathogenesis of AD.The purpose is to study the influence of calculation method and Post label delay(PLD)parameter selection on CBF and the relationship between CBF and cognition in different course of AD during clinical scanning.Conventional MRI sequences and 3D-ASL sequences with PLD=1500ms and PLD=2000ms were performed on 47 subjects(16 healthy subjects,15 mild subjects and 16 severe subjects)using a 3.0T high magnetic field magnetic resonance imager(GE DISCOVERY MR750).Cerebral perfusion images were generated by using the built-in computing system of magnetic resonance apparatus and ASLtbx toolbox,and standard spatial cerebral blood flow images were generated by using SPM in MATLAB.Use the harvard-Oxford cortex and cortex structure atlas as a template to get the subjects cover gray matter of regional cerebral blood flow in 112 regions(of regional cerebral blood flow,rCBF)data,using SPM,SPSS,XJview and Graphpad software for the data of one-way ANOVA,covariance analysis,multiple comparison correction and to show the result.We found that cerebral perfusion images generated by the MRI system and the parameter selection of PLD=2000ms could better reflect cerebral perfusion.Cerebral blood flow in the patient group was significantly lower than that in the healthy group.In the voxel-based analysis method,perfusion in 27 brain areas in the severe group was significantly lower than that in the healthy group.In the analysis of regions of interest,34 brain regions in the severe group were found to be significantly lower than those in the healthy group.The right angular gyrus and the right posterior cingulate gyrus of the 34 brain regions were positively correlated with MMSE.This study not only provides a method for clinical cerebral blood flow research,but also explores the role of local cerebral blood flow changes in patients with AD.To a certain extent,it can reflect the status of cognitive function and has the possibility of becoming a biomarker,which needs further research and discussion.
