Liver is an important organ for lipid metabolism. In addition to CDP-choline pathway for phosphatidylcholine(PC) biosynthesis, there was a special pathway catalyzed by phosphatidy -lethanolamine-N-methyltransferase2 ( PEMT2 ) which add three methyl groups to phosphatidylethanolamine (PE). In the lipid metabolism of liver, there were always some parts of phospholipids turn to plasmalogen forms which only account for 0~5%. A question that deserves to be ask here is: "Is there any special physiological function of plasmalogens on the signaling pathway that leads to cell proliferation in the liver?"Plasmalogens are a unique subset of phospholipids in which the sn-lcarbon of the glycerol back bone contains a vinyl ether-linked (a cis double bond adjacent to an ether bond) long chain hydrocarbon instead of the typical ester-linked fatty acid. Plasmalogens are widely distributed in mammalian tissues. The major classes of plasmalogen are plasmenylethanol -amine (PlasEth or plas-PE) and plasmenylcholine (PlasChol or plas-PC). Different tissuses and possibly even different cell types within one type of tissue, may have variable amount of plasmalogens. The physiological role of plasmalogens is not fully understood yet so far, putative ructions for plasmalogens may include the ability to serve as endogenous antioxidants, their possible role in membrane dynamics, and as storages of PUFAs and lipid mediators, In addition, synthetic plasmalogens have selective toxin on cancer cells which drawing notice of more and more researches.to this study, plas-PE and plas-PC derived from the liver of pig were extracted and purified. By using MTT assay and flow cytometry (FCM), we observed the inhibition of plasmalogens on the growth of rat hepatoma cell CBRH-7919 and other cell lines .To further elucidate the mechanism of plasmalogens on proliferation of the hepatoma cells, western bolt assay were performed to identify expression of the signaling proteins. Furthermore the molecular mechanism of inhibition of proliferation induced by transfection of pemt2-cDNA into rat hepatoma CBRH-7919 cells was unraveled. The aim of this study is to understand the effects of signaling lipids on biology function of liver and proliferation of hepatic cells, and offer theoretical evidence and technological support for developing natural antitumor phosphoslipids drugs.(1) The total phospholipids were extracted and PC and PE were separated, plasmalogens were purified by method of high performance thin layer chromatography (HPTLC) with different...
