Plasmalogens are a subclass of phospholipids in which the sn-1 position has a long chain alcohol attached through a vinyl ether bond. Plasmalogens are widely distributed in mammalian tissues, and account for ten percent of the total phospholipids. The major classes of plasmalogen are plasmenylethanolamine (PlsEth or pls-PE) and plasmenylcholine (PlsChol or pls-PC). As the chemical properties and the distribution in cell membrane, plasmalogens play an important role in cell membrane fusion, transportation of ion and cholesterol, as well, signaling as a reservoir for the second messengers. Especially, the roles of plasmalogens in anti-tumor and anti-oxidization are drawing notice of more and more researches. In this paper, the phospholipids and constituents of fatty acid in livers from different animals were analysed, comparatively and respectively. Plasmalogens were extracted and purified, and the effects of plasmalogens on proliferation of rat hepatoma cell CBRH-7919 were observed. (1) The phospholipids and constituents of fatty acid in livers from fowl, rabbit, sheep and swine were analysed comparatively by using the methods of organic reagent extracting, Folch fraction, HPTLC and GC. The result shown that the phosphatidylcholine(PC) and phosphatidylethanolamine(PE) were the major types of phospholipids. There is no significant difference on the composition of phosphlipids among the livers from different animals. Thus, the livers from the four different animals would become the sources of plasmalogen. (2) The total phospholipids were extracted and PC, PE were separated as above, non-plasmalogens and plasmalogens were separated and purified by the method of HPTLC with different developing system. (3) The rat hepatoma cell CBRH-7919 were cultured and the effects of plasmalogens on proliferation and apoptosis of CBRH-7919 cell line were observed by the methods of MTT test and flow cytometry. The results shown that the plasmalogens can inhibited the proliferation of CBRH-7919 cells, obviously and dose-dependently. At the same time, some cells went on apoptosis, which suggested that the inducing apoptosis may be involved in the mechanism of anti-tumor of plasmalogens.(4) The mouse hepatoma cell H22 were transplanted into mice and the effects of different dose of plasmenylethanolamine on proliferation of H22 cell were observed by measuring tumor weight and volume, and calculating the rate of inhibition. The results shown that different dose of plasmenylethanolamine can inhibited the proliferation of H22 cell obviously, which indicated that plasmenylethanolamine can be apply to clinical therepy.
