Abstract/Summary:
Objectives:To investigate the apoptosis effect of gastric cancer SGC-7901 treated by curcumol, inhibition ratio of proliferation of gastric cancer SGC-7901 cells, morphology changing, DNA changing and expression of important apoptosis protein Bcl-2 were detected. The results will be helpful for elucidating mechanism of curcumol anticancer, offering experimental data for clinic applying.Object:Human gastric cancer cells line: SGC-7901.Methods:Curcumol was dissolved in dehydrated alcohol and initial concentration of curcumol was 5mg/mL. Group A,B,C and D as experimental groups were set up .The curcumol concentrations were set as follows: 12.5,25,50,100μg/mL. Group E was control group, containing 1% dehydrated alcohol. After seeded for 24, 48 and 72h, inhibition ratios of SGC-7901 cells proliferation were detected by MTT assay and alamar blue assay, and morphology changings of apoptosis were detected by AO/EB fluorescent staining. After seeded for 24 and 48h, apoptosis ratios were detected by FCM and the expressions of bcl-2 were detected by Western Blot.ResuLts:1. 12.5-100μg/mL curcumol could restrain proliferation of SGC-7901 after treated for 24, 48 and 72h detected through MTT assay.
