Objective This study was designed to evaluate the spermatogenic cellsapoptosis and the expression of Bcl-2 and Bax gene in the spermatogenic cells of smoking rats, and to explore the corelation between spermatogenic cells apoptosis and the expression of Bcl-2 and Bax.Methods Fifty male Sprague-Dawly rats( 60 days old) were dividedrandomly into five groups, the animals were exposed by smoke in 6.49g/m~3(A), 12.98g/m~3(B), 25.96g/m~3(C), 12.98g/m~3(D), 0g/m~3(E), for 30min twice a day, were passivity breathed cigarette smog for 4 weeks. The rats were sacrificed and weighed the testis, the morphology of spermatogenic epithelium in rats of every group were observed with light microscopy. The cell apoptosis was detected by TUNEL method, and using streptavidin-biotin peroxidase (S-P) method to detect the expression of Bcl-2 and Bax protein.Results The weight of testis were diminution by the increased of cigaretteamount in the smoking group, and there are statistically significant difference among the groups. Bax in the smoking group are highly expressed than in the comparison group, the difference is significant. On the other hand, there is no statistically significant difference for Bcl-2 between smoking group and comparison group. A significant correlation between bcl-2, Bax gene expression and spermatogenic cells apoptosis in smoking group rats.Conclusion1. Smoking could induce spermatogenic cells apoptosis and oligozoospermia, and Smoking could induce diminution of the weight of rats testis. To abnegate smoking might be benefited to the renewing of spermatogenic function.
