Chemokine SDF-1α and its receptor CXCR4 plays an important role in embryogenesis, tumor metastasis and mediation HIV infection. Latest SDF-1α /CXCR4 is discoveried to be highly expressed in atherosclerosis plaque, so its role in atherosclerosis has been highlighted.In our study, the effect was explored of SDF-1α /CXCR4 on THP-1 chemotaxis firstly. Then SDF-1α /CXCR4 expression were researched in rVSMC and THP-1 cells pretreated with Ox-LDL. Then the role of SDF-1α /CXCR4 was evaluated in VSMCs/monocytes adhesion.Objective To explore the effect of SDF-1α /CXCR4 on THP-1 cells chemotaxis and influence of Ox-LDL, momordicin on THP-1 cells migration exposed to SDF-1α , the effect of Ox-LDL on the expression of CXCR4 in THP-1 cells and SDF-1α in cultured primary rVSMC, the effect of SDF-1α /CXCR4 on THP-1/rVSMC adhesion. Methods THP-1 cells chemotaxis was examined with transwell permeable supports. THP-1 cells with or without pretreated with Ox-LDL were input in upper chambers, SDF-1α and momordicin were added in upper and/or low chambers. Number of migrated cells in the low chamber was counted after incubation for 10h. CXCR4 or SDF-1α mRNA and protein was revealed by RT-PCR and Western blot respectively in THP-1 cells or rVSMC incubated with different concentrations/time of Ox-LDL. SDF-1α expression was examined by immunohistochemistry in rabbit model with common carotid artery ringer. rVSMC pretreated with 50μg/ml Ox-LDL for 48h were incubated with THP-1 cells and with or without SDF-1α antibody to observe cells adhesion.Results SDF-1α induced a pronounced migration of input cells in a concentration-related manner, but this effect was obviously inhibited by the antibody to CXCR4.
