Tibetan Hand-parametric Pharmacodynamic Experimental Study

1.PurposeTuber Gymnadeniae(Shoushen),also called Taipingshen,is the common-used Tibetan herbal medicine.It has the actions of tonifying original Qi and nourishing kidney essence,strengthening the body,tranquilizing mind and improving intelligence,and prolonging life span.Tuber Gymnadeniae is effective for the treatment of insomnia,weakness after being illness, digestive ulcer,and chronic nephritis.Till now,there are less research reports about Tuber Gymnadeniae.So it is necessary to carry out further pharmacodynamic research on the main therapeutic effect of Tuber Gymnadeniae, which will supply evidence for the extension of its clinical use.2.Methods and Results2.1 Experimental study on sedative and hypnotic actions of Tuber GymnadeniaeWith normal saline as the control,the sedative and hypnotic actions of Tuber Gymnadeniae at different dosages were observed.The results showed that Tuber Gymnadeniae had an effect on reducing spontaneous activity of mice,decreasing the upward-raise frequency of the mice forelimbs,prolonging sleeping time of mice induced by pentobarbital sodium over threshold dosage and increasing the number of sleeping mice induced by pentobarbital sodium less than threshold dosage.It is indicated that Tuber Gymnadeniae has obvious sedative and hypnotic actions in a dose-dependent manner,and its mechanism need further research.2.2 Experimental study on kidney-yang-tonifying action of Tuber GymnadeniaeThe results showed that Tuber Gymnadeniae relieved the symptoms of yang deficiency in mice induced by hydrocortisone,increased the body weight kidney-yang-deficiency mice,prolonged retention time on the rotating bar, and increased mice kidney index,the difference being significant as compared with the model group.Tuber Gymnadeniae could also increase the splenic and renal DNA synthesis rate,the difference being significant.It is indicated that Tuber Gymnadeniae has the actions of tonifying kidney and strengthening body on mice with kidney-deficiency induced by hydrocortisone. 2.3 Experimental study on Tuber Gymnadeniae for the preventing and treating gastric ulcerWith normal saline as the control,the inhibitory effect of Tuber Gymnadeniae on gastric ulcer induced by the mixture of hydrochloride-ethanol was observed.The macroscopic and pathological results showed that Tuber Gymnadeniae relieved gastric ulcer,decreased serum and gastric MDA content in rats.It is indicated that Tuber Gymnadeniae has a certain inhibition on gastric ulcer induced by the mixture of hydrochloride-ethanol,and can decrease the serum and gastric MDA content.3.DiscussionTuber Gymnadeniae has obvious sedative and hypnotic actions,can tonify kidney and strengthen body on mice with kidney-deficiency induced by hydrocortisone,and exerts protective effect on gastric ulcer induced by the mixture of hydrochloride-ethanol.Its therapeutic mechanism may be related with increasing gastric mucosal activity,decreasing MDA content and reducing gastric mucosal injury induced by free radicals.
