Objective: To detect the expression of Survivin and Bcl-2 gene and apoptosis index (AI) in cholangiocarcinoma and to analyze its relationship and the relationship with clinicopathologic characteristic in cholangiocarcinoma.Methods: Immunohistochemical SP method was used to detect Survivin and Bcl-2 gene expression in 52 cases of Cholangiocarcinoma and 34 cases of the normal or benign lesion bile vessel tissue. The cell apoptosis of 52 cases of cholangiocarcinoma was detected by TUNEL method. The AI in Survivin and Bcl-2 positive and negative expressional groups were calculated. Results: (1)The positive rate of Survivn expression in cholangiocarcinoma was 65.38%(34/52) and that in normal or benign lesion bile vessel tissue was 0%(0/34).There was statistical difference between them(p<0.01). (2)The positive rates of Bcl-2 expression in cholangiocarcinoma and normal or benign lesion bile vessel tissue were 44.23%(23/52),8.82%(3/34),respectively. There was statistical difference between them(p<0.01). (3) There was not a significant difference among the expression of Survivin protein in different sex,ages,tumor location,histodifferentiation and TNM stages of the cancer. However there was a relationship between Survivin expression level and lymphatic metastasis. (4) There was not a significant difference among the expression of Bcl-2 protein in different sex,ages,tumor location,lymphatic metastasis and TNM stages of the cancer. However there was a relationship between Bcl-2 expression level and tumor histodifferentiation. (5)There was significant positive correlation between Survivin and Bcl-2 expression(γ=0.529). (6)The AI in Survivin positive and negative expressional groups were 0.55±0.25%,1.62±0.30%,respectively. There was statistical difference between them (p<0.01). The AI in Bcl-2 positive and negative expressional groups were 0.65±0.34%,1.71±0.39% ,respectively.There was statistical difference between them(p<0.01).Conclusions:(1) Survivn was highly expressed in cholangiocarcinoma and the expression of Survivin was associated with lymphatic metastasis. (2)Bcl-2 was highly expressed in cholangiocarcinoma and the expression of Bcl-2 was increased with the heightening differentiated degree. The lower expression of Bcl-2 gene can be one of detections of poorly differentiation and unfavourable prognosis of cholangiocarcinoma. (3)The cell AI of Survivin and Bcl-2 negative expressional groups in cholangiocarcinoma were obviously higher than that of positive negative groups. (4)Survivin gene expression was positive correlated with Bcl-2 gene expression.
