Background/Purpose: In this experiment we used somatostatin anologue octreotide to investigated it's growth inhibition effect on cholangiocarcinoma cell QBC939 in vitro and in vivo and probile mechanisms..Method: Cholangiocarcinoma cell line QBC939 were cultured in vitro. MTT assay were used to determineted the antiproliferative effect of different doses somatostatin anologue octreotide on cholangiocarcinoma cell QBC939 and growth rate were calculated. Cell cycle in the cholangiocarcinoma cell QBC939 after dfferent doses somatostatin anologue octreotide administration were evaluated by flow cytometry. Effect of OCT on the expression of p27kip1 were evaluated by immunochemical. Nude mouse tumour heterograft model were established and used to evaluat antiproliferative effect.Result: MTT assay showed that OCT significantly reduced QBC939 cell growth and were dose-dependent.After treated by 0.005~5mg/L OCT for 48 hours,the cell growth rate decended gradually,respectively 83.8% 80.28% 77.53% 65.32%.Between control group and experimental group the OD value was significant different (p< 0.05) . Result of flow cytometry showed that OCT can effect the location of cell cycle. It could make cell cycle arrested in G0/G1 phase. Expression of p27kip1was depended on doses of octreotide .OCT can increase it's expression .The weight of QBC939 xenografts in OCT-treated group were lower than those in the control group.Conclusions: The result proved that OCT inhibits the proliferation of human extrahepatic cholangiocarcinoma cell line QBC939 .The mechanism of it's effect was cell cycle arrested. The up-regulation of p27kip1 might be correlated with cell cycle arrested...
