Objective To set up endometriosis model in rats and explore the role of aromatase in endometriosis. Comparison with Nemestran(Gestrinone) and Anastrozol(Aromatase Inhibitor), to observe the effects of Luoshi Neiyifang on aromatase expression of ectopic and eutopic endometrium in rats with endometriosis.Methods Self-endometrium transplantation by surgical method was used to develop rats models of endometriosis. They had been raised by Luoshi Neiyifang, Nemestran, Anastrozol respectively for 3 weeks after 4 weeks of modeling. Radioimmunoassay was used to determine the level of E2, TXB2, 6-Keto-PGF2α in serum and fluorescence quantitative PCR(FQ-PCR) was used to detect the expression of aromatase in ectopic and eutopic endometriums in rats.Results (1)The livability of replanting on the ovary of rats model was 83. 3%. (2) The diameter of the ovarian endometriosis cysts in all the medication groups were less than those in non-medication groups(p<0.05). (3) The expressions of aromatase in eutopic endometriums of model-free group were lower significantly than those of model groups (p<0. 05). The expressions of aromatase in eutopic endometrium were lower significantly than those of ectopic one (p<0. 05). All the medicines in this experiment can inhibit the expression of aromatase in different degrees. The prevalence rate was significant different between the control group and the groups includingAnastrozol, Nemestran(the low concentrations one)and Luoshi Neiyifang(the high concentration one) (p<0. 05). But in eutopic endometrium the values of the heavy Nemestran group were the highest among the medication groups(p<0.05). (4)The high concentration Luoshi Neiyifang and Anastrozol (both high and low concentration included) can decrease the level of E2 in serum significantly (p<0.05), but those were not as many as the Nemestran ones. (5)Compared with Nemestran, Anastrozol, treatment-free groups, Luoshi Neiyifang(the high concentration group) can decrease the level of TXB2 in serum and the value of T/K.Conclusion Establishment of endometriosis model by autologous transplantation on SD rats during non-estrus with estrogen was success and credible, which was especially suitable for large sample research. Aromatase plays an important role in endometriosis. The way through which activity of aromatase is inhibited and estrogen is decreased may be applied. Luoshi Neiyifang with certain concentration may inhibit the expression of aromatase and decrease E2 of serum. But the advantages of the Chinese medicine are also showed as avoidance of estrogen with low level and stability of microcirculation.
