AIM:â‘ To investigate the expression of cancer-testis (CT) antigens SSX1-5 mRNA in meningioma and hepatocellular carcinoma (HCC) and the possibility of applying these antigens as targets for specific immunotherapy for meningioma and HCC.â‘¡To clone a fragment of SSX4 gene into an expression vector pMAL-C2 for expressing SSX4 protein in vitro.METHODS:â‘ Expression levels of CT antigens SSX1-5 mRNA were detected with reverse transcription polymerase chain reaction (RT-PCR) in 69 meningioma tissues, 61 HCC tissues and 52 HCC corresponding adjacent tissues. Correlation between the expression of CT antigens SSX1-5 and clinical data was evaluated.Genes of positive PCR results were sequenced randomly.â‘¡Using testis cDNA as template , amplified the KRAB fragment of SSX4 gene with RT- PCR.Insert this fragment into the expression vector pMAL-C2, and identified the positive clone with white and blue assay,endonuclease, PCR and DNA sequenceing identification.RESULTS:â‘ Of 69 meninginoma tissues, SSX1 and SSX2 mRNA were detect- able in 1.5% (1/69)meningioma samples, respectively. Non of the others was detectable.Of 61 HCC tissues, SSX1, SSX2, SSX4 and SSX5 mRNA were expressed in 42.6% (26/61), 8.2% (5/61), 39.3%(24/61) and 6.6% (4/61), respectively.About 50.8% (31/61),32.8% (20/61),9.8% (6/61) and 3.3% (2/61) of HCC tissues positively expressed at least one, two, three, and four members of SSX1,SSX2,SSX4 and SSX5,respectively.In addition,non of 52 samples of HCC corresponding adjacent tissues expressed SSX1-5.
