Rheumatoid arthritis (RA) is one of autoimmune diseases which pathogenesis has not been elucidated. The animal model with collagen-induced arthritis(CIA) was established in Wistar rats with CCII in CFA by intradermal injection and intervened by antisense oligodeoxynucleotide of MMP-2 and MMP-9 to investigate the intervened effect .It was immuned by intradermal injection of CCâ…¡emulisified with complete Freund's adjuvant(CFA). The pathological characteristics of CIA were assayed by observing the swelling of anklebone and toes,hair color and action state,the level of anti-CCII antibody in serum. The soft swelling and the articular bone erosions were analyzed by roentgenography and histopathology techniques, respectively. The results showed obvious swelling of the anklebone and toes in the experiment group compared with the normal group. Anti-CCII antibody level was higher in experiment group than that in the normal group by ELISA(P<0.001). Besides, typical arthritis pathology was showed by roentgenography and histopathology.It was divided into three groups with different dosage for intervene: 1.037mg/kg,8.333mg/kg and 33.33mg/kg. After injection, the state of rats got better by observing the swelling of anklebone and toes,hair color and action state. Gelatin zymography-PAGE results indicated that the inhibition effect in the group injected with dose of 8.333mg/kg and 33.33mg/kg was better, and the inhibition effect in the group injected with dose of 1.037mg/kg was not obvious.
