objective To observe the effect of transplantation of the marrow stroma cells (MSCs) modified with gene of NT4 in hippocampus of AD rat on the behavior, neuroelectrophysiology and morphology.Methods Thirty femina SD rats were randomly divided into five groups: normal group; AD model group; DMEM transplantation group; MSCs transplantation group and MSCs-NT4 transplantation group. AD models were made by transecting bilateral fimbria-fornix. On the tenth day after surgery, the MSCs, MSCs -NT4 and DMEM were separately transplanted into the hippocampus of AD rats. One month after transplantation, the behavior tests were carried out. When then the behavior tests were over, the electrophysiology tests were carried out. The rats were killed when all tests were finished. Brain sections were processed by immunohistochemical procedure with molocloned antibodies against ChAT. Number of ChAT positive neurons were counted and analyzed statistically. Gene products of NT4 were tested by immunohistochemical procedure with molocloned antibodies against NT4. Results 1. Behavior tests showed that the average of escapelaterncy of MSCs and MSCs-NT4 transplantation groups all obviously shorten compared to AD group (P< 0.01); There was no difference between AD group and DMEM group (P > 0.05 ); The average of escapelaterncy of MSCs-NT4 group shortened compared to MSCs group (P<0.05 ). 2. neuroelectrophysiology P50 test: The amplitude of C-P50 and T-P50 of AD group obviously reduced compared to normal group (P < 0.01); After...
