Objective: To investigate the effect of imiquimod on the miniature pig's back scarmodel and fibroblast of pathological scar about its biological behavior andpathological changes.Methods: The fibroblasts of scars were cultivated in vitro, scar models were made onminiature pig's back. The biological behavior and pathological changes of thefibroblasts and local immunological modulation affected by imiquimod wereobserved and analyzed by inverted microscope, MTT assay, cells' and tissue's HEstaining, Masson's improved staining, immunochemical staining and transmissionelectron microscope(TEM).Results: In vitro: by MTT assay and cells' HE staining, the growth and proliferationof fibroblast of experiment groups were decreased compared with the control groups.In vivo (miniature pig's back scar model): by pathological and immunologicalstaining and TEM, the proliferation of the fibroblasts were decreased compared withthe control groups, at the same time the collage's decrease was increased comparedwith the control groups.Conclusions: Imiquimod can decrease fibroblasts growth and proliferation in vitro.5% imiquimod cream can increased the mount of cytokine, such as INF, TNF, IL, et al,and induced the imbalance of MMP-1/TIMP-1 in scars, activated collagenase andincreased collage's decrease in vivo; it also can decreased Bcl-2 and increasedapoptosis of the fibroblasts, decreased the mount of fibroblasts and collage synthesisin vivo.
