| Objective:To investigate the nephroprotective effects of ShenKangling on patients with chronic renal failure(CRF) and rats with 5/6 nephrectomy and explore the machenism of.Methods:clinic study:68 patients with CRF were divided into 2 groups:38 treated with Shenkangling and 30 treated with benazepril,who were treated for 8 weeks.Urinary protein excretion for 24 hours,blood urea nitrogen (BUN),serum creatinine (Scr),creatinine clearance (Ccr),hemoglobin(Hb),plasma albumin(ALB),triglyceride (TG),cholesterol (CHO),blood rheology and transforming growth factorβ1(TGFβ1) in serum were tested after treatment.Experimental reseach:50 wistar rats were randomly divided into 5 groups:Sham group,5/6 nephrectomized group,benazepril group (10mg·kg-1·d-1),ShenKangling high-dosage group (15mg·kg-1·d-1) and ShenKangling low-dosage group(5g·kg-1·d-1).Urinary protein,BUN,SCr and ALB were measured after 8 weeks at the end of the study.Glomerulosclerosis index(GSI) were evaluated by PAS and HE stain.Immunohistochemistry was used to examine the expression of fibronectin(FN),laminin(LN),collagenⅣ(COL-Ⅳ) and TGFβ1.Results:clinic study:the efficiency treated with Shenkangling and benazepril were 84.20% and 83.3%,respectively.Urinary protein excretion for 24 hours,BUN,Scr,Ccr and ALB were improved by Shenkangling and benazepril equally.The expression of TGFβ1 in serum was greatly decreased as well as blood rheology improved by Shenkangling,while Shenkangling was better in increasing Hb.Experimental research:Compared with the untreated 5/6 nephrectomized group,Urinary protein,BUN and SCr of ShenKangling group and benazepril group were...
