Aim Yichangping(YCP) is a civilian complex prescription of Chinese herbal medicine which is commonly used for treating ulcerative colitis(UC) and often gives surprise therapeutic effect. This study was aimed to determine the efficacy of YCP and to find out its mechanism in two experimental models resembling human UC.Mothods UC models in mice were induced by using 2% DSS and acetic acid respectively, and then the mice were divided into six groups randomly, 12 mice in each group, namely blank control group , model control group , solfasalazine-treated group(SASP , 200 mg·kg-1·d-1),and YCP high(YCP1, 30.8 mg·kg-1·d-1), medium(YCP2, 15.4 mg·kg-1·d-1), low(YCP3,7.7 mg·kg-1·d-1) dosage group. Both blank and model control groups were given partes aequales distilled water instead of the drugs. (1)Disease active index (DAI) and macroscopic score of colonic damage(MSCA) were detected daily. The numbers of total white blood cells(TWBC) and the percentages of neutrophile(N%) were measured.(2)The permeabilities of colonic mucosa(PCM) were measured by evens blue method. Immunohistochemical staining method was used to determine the expressions and the distributions of iNOS and NF-Κbp65 in colon. The activities of total superoxide dismutase (SOD) and malondialdhyde (MDA) level in colonic tissue were measured by xanthine oxidase method and thibabituric acid (TAB) method respectively. The levels of TNF-αwere also measured by enzyme linked immunosorbent assay(ELISA) method.(3) Maximally tolerated dose (MTD) in mice was determined.Result 1.DAI and MSCA: The DAI in blank control group, model control group, SASP treated group, YCP1, YCP2 and YCP3 group were 0.00±0.00, 6.33±2.90, 3.16±1.75,...
