Objective: To investigate the relationship between transforming growth factorβ1(TGFβ1), basic fibroblast growth factor(bFGF) and the detrusor underactivity following BOO in the rat, to probe the changes of TGFβ1 and bFGF in the detrusor after PBOO on the level of mRNA, to investigate the expression and clinical significance of TGFβ1 and bFGF levels in the rat urine after PBOO. To discuss wether the two factors participate the pathophysiology of bladder dysfunction due to BOO.Materials and Methods:1. To set up the model of PBOO in the Wista rat: 30 Wistar rats aged 3 months arerandomly divided into three groups, raised in separate cage. They are respectively Sham operation group, 8 rats. PBOO 2 week group, 11 rats whose BOO are maintianed 2 weeks. PBOO 6 week group, 10 rats whose BOO are maintianed 6 weeks. Proximal urethral ligation methods are used to establish Wistar rats PBOO models.2. Excised bladder weight are measured after successful models.3. Detrusor contraction experiments in vitro stimulated by Carbamylmethylcholine(Carbachol): Wistar rats are executed the respectively after 2 weeks and 6 weeks of PBOO.
