AIM: To investigate the preventive and therapeutic effects of dog bile on alcohol-related hepatitis in rats.Methods: Rat were administered ethanol (40% v/v) dosage of 8g/kg body weight daily by gavage for four wks, then the dosage of ethanol (45% v/v) was enhanced to 9g/kg for four wks. At the same time, the rats in dog bile group received 400mg/kg of dog bile solution orally once a day for 8 wks. The activities of serum of alanine transaminase (ALT) ,aspartate transaminase (AST), γ-GT values, and the ratio of liver/body weight were detected. Meanwhile, the pathological changes of livertissues were observed under light microscope.The expressions of α-smooth muscleactin( α -SMA) was detected by immunohistochemical SP method and TNF- α wasdetected by LAB-SA method.Results: Compared with that in the model group, the levels of ALT, AST and γ-GT were significantly decreased,and the ratio of liver/body weight was increased in the dog bile group.The pathological changes were lighter,and only a little collagenousfiber was observed, the expressions of TNF-α and α-SMA in liver were reduced inthe dog bile group.Conclusion : The dog bile can prevent the alcohol-related hepatitis in rat, themechanism may relate to its inhibition the TNF-α secretion by Kupffer cell and abatment liver injury.
