Study On The Mechanism Of Supplementing Qi And Activating Blood Circulation To Prevent And Cure Atherosclerosis (AS)

Object:(1)To form high-fat-diet rabbit model of aorta atherosclerosis.(2)To reveal the action mechanism of complex prescription of Traditional Chinese Medicine , which has the function of supplementing qi and activate blood circulation, to prevent and cure atherosclerosis.Methods: Form the AS animal model caused by deficiency qi and blood stagnation with high cholesterol diet and movement tiredness etc.. The rabbits were administrated with the complex prescription of TCM intragastricly as forming the model in TCM prophylaxis group. After successive 8 weeks, the animals were fed on the feedstuff from which the cholesterol was eliminated for 4 weeks. During these successive 12 weeks,. the rabbits were divided into TCM complex prescription group administrated with TCM, control group of western medicine administrated with Lipanthyl solution intragastricly and other groups administrated with sodium chloride. Then detect NF-κB, CRP, IL-6, VCAM-1, TC, TG and LDL-C in the vein blood sample of ear fringe, and take aorta to do the sample of electron microscope and light microscope. Results:The TC of model group was higher than the normal group as well as TG, LDL-C and the ratio of aorta plaque to endothelium area (P<0.01), however the TCM group was lower than the model group (P<0. 01). The results of immunohistochemistry and image analysis indicated that expression of NF--κB and VCAM-1 in model group rabbits was promoted (P<0. 01), but they were decreased in TCM group(P<0.01). Compared with the normal group, CRP and IL-6 were higher conspicuously(P<0.01), however they were lower in TCM group(P<0.01). Conclusions:The mechanism of complex prescription of Traditional Chinese Medicine, which has the function of supplementing qi and activate blood circulation, to prevent and cure atherosclerosis was as the following:1. Regulating the lipid metabolism: decrease TC, TG, LDL-C.2. Inhibit the expression of NF-κB and VCAM-1 so as to prevent and cure AS.3. The generation process of AS may be influenced by NF- κB, CRP, IL-6 and VCAM-1 importantly.
