Dendritic cells(DCs) orchestrate immune response and may also contribute to the pathogenicity of autoimmune disease systemic lupus erythematosus(SLE). The deviation of DCs may partly be explained by the abnormal cytokines, for example, the inhibitory molecule IL-10. In order to study the roles of SLE serum on the DC in the SLE, we randomly collected 56 SLE patients and 9 normal controls to analyze the levels of IL-10 in serum by ELISA. Normal PBMCs were separated and monocytes were cultured by GM-CSF and IL-4 in order to differentiate into DCs. In the process, we added SLE patients serum with different level of IL-10. DCs were identified by morphology, molecular expression and mixed leukocyte reactions (MLR).Also,the DCs were further cultured for 2 days and supernant was evaluated with IL-10, IL-12p40, and IFN-γ. Result showed that high IL-10 level in SLE serum downregulated the expression of HLA-DR and CD80 and seemed no effect on CD86, after neutralization of IL-10 in SLE serum with antibody to IL-10, the expressions of HLA-DR and CD80 were restored. Whatever the reduced expression of HLA-DR and CD80, DC induced in SLE serum combined with GM-CSF and IL-4 had much lower stimulatory ability in MLR compared with normal controls.
