Objective: To investigate the relationship between pulmonary arterial wall collagen and pulmonary hypertension when pulmonary hypertension induced by high pulmonary blood flow happen.To observe the effects of lowering pulmonary arterial pressures and pulmonary arterial wall collagen by simvastatin and combined use of simvastatin and enalapril.Method:32 male Sprague-Dawley rats were randomly divided into splitless group(n=8),shunting group(n=8),solo-medication group(n=8) and combined-medication group(n=8).Abdominal aorta and inferior vena cava shunting were produced in rats of the later three groups.The rats of solo-medication group were fed by simvastatin with dose of 2mg/kg.d to interfere and the rats of combined-medication group were fed by simvastatin with dose of 2mg/kg.d and enalapril with dose of 15mg/kg.d to interfere.All rats in four groups were fed in identical niche for 11 weeks.Pulmonary artery systolic pressure(PASP) of each rat was measured by using a right cardiac catheterization procedure after 11 weeks.At the same time,blood in abdominal aorta and lung tissue of each rat were drawn from it.Pulmonary vascular microstructure was measured.The ratio of vascular wall thickness/vascular external diameter(WT%)and the ratio of vascular wall area/total vascular area(WA%) were documented. AngiotensinⅡ(AⅡ) content of blood and lung tissue was measured by spectrophotometry.The expressions of collagenⅠ,Ⅲand transforming growth factor-β1(TGF-β1) in pulmonary arterial wall were semiquantitative by immunohistochemistry and image analysis.
