Objective To explore the changes of peripheral blood neutrophil apoptosis and its role of survivin expression on severe acute pancreatitis (SAP) .Methods Seventy-two Sprague-Dawley(SD) rats were randomly divided into two groups: SAP group and sham operation (SO) group.The SAP group was fourty-eight rats, and the SO group was twenty-four . The model of SAP was induced by retrograde injection of 4% sodium taurocholate(1ml/kg) into the bili-pancreatic duct in rats. Rats in the two groups were killed at 24,48,72hours after induction of the model. The serum amylase level and the histological pathologic changes of pancreas were measured at different time point in each group. The peripheral blood neutrophil was isolated and its apoptosis were measured by TUNEL staining methods. The expression of peripheral blood neutrophil survivin protein and mRNA were determined by immunohistochemistry and semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR), respectively.Results (1)The levels of amylase in SAP group increased significantly compared with those in SO group(P<0.05);(2)The pancreatic histopathologic scores in SAP group increased significantly compared with those in SO group, and tended to rise gradually with time prolonged(P<0.05); (3) Neutrophil apoptosis rates in SAP group decreased significantly compared with those in SO group, and tended to degrade gradually with time prolonged(P<0.05);(4)Survivin protein and mRNA in the peripheral blood neutrophil from SAP group increased significantly compared with those from SO group, and tended to rise gradually with time prolonged(P<0.05);(5)The pancreatic histopathologic scores was negatively correlated with apoptosis of neutrophil in SAP group (r=-0.97, P<0.05)(;6)Survivin protein and mRNA expression were negatively correlated with apoptosis of neutrophil in SAP group, respectively (r=-0.97,and r=-0.99, P<0.05).
