Objective To further vertify the bioactivities and mechanism of piperine and to investigate the effect of piperine on the ion channel of insect cytes, the toxicity and lethal effect of piperine against Culex pipiens pallens larvae and Spodoptera frugiperda cell line were investigated under the laboratory conditions.Methods The LC50 of 4th-instar larvae of Culex pipiens pallens was evaluated.Poisoning symptom and lethal effect was observed. Modified Giemsa stain was used to observe the effects of piperine on the morphology alteration of Culex pipiens pallens larvae lymph cells. The toxicology of piperine on Culex pipiens pallens larvae was investigated.Using MTT-colorimetric, Modified Giemsa stain to observe the effects of piperine on the mortality and morphology alteration of Sf-21cells. Trypan blue dye exclusion assay was used to observe the effects of piperine on the growth rate and restoration of Sf-21cells and to reveal the toxic effect and growth inhibitory of piperine on Sf-21cell line.Results The main results are given as follows: (1) Piperine showed strong contact activity against the 4th-instar larvae of Culex pipiens pallens, whose values of LC50 was 0.368μg/L. (2) Typical poisoning symptom of piperine was:excitation,spasm, exanimation and ultimate death.
