Objective: To investigate the expression of NF-κB and OPN in the tissue of diabetic nephropathy, and relationship between the pathogenesy of immunology in diabetic nephropathy and the expression of NF-κB and OPN, and the intervention effect of mycophenolate in diabetic nephropathy.Methods: In the study, healthy male Sprague-Dqawiey rats 51 weighing about 200g were randomly divided into three groups: 15 rats were divided into the group normal control (group A), 36 of which was received a single intraperitoneal injection of STZ dissolved in 0.1mol/L sodium citrate (pH 4.5) at a dose of 55mg/kgWT , which of the blood glucose exceeding 16.65mmol/L were divided into group B (diabetic group) and group C (mycophenolate -treated group) which was treated with mycophenolate (10mg/kg/d, 5mg/ml), and normal saline was given to the other groups. The rats for every group were respectively sacrificed at week 8, and week 12 after success induced. Urine samples were collected before sacrificed and blood-serum samples were separated from heart. The expression of NF-κB,OPN,TGF -β1 proteins was evaluated by immunohistochemistry.Results: 1.The level of blood glucose, ratio of renal weight/body weight and urine protein increased progressively in all group B and group C are higher than group A significantly(P<0.05). 2.pathological section indicated that pathological changes of diabetic nephropathy was intensified as course of disease prolonging. 3.The expression of NF-κB in glomcrulus and nephric tubule at 8 week in gruop B were 7.91±3.96, 21.08±8.25 respectively, which were higher than group A significantly(P<0.05). The expression of NF-κB in glomcrulus and nephric tubule at 8 week in group C were lower than group B significantly(P<0.05),but higher than group A significantly(P<0.05).
