The Effects Of Puerarin And Daidzein On Calcium Metabolism In Ovariectomized Rats

Puerariae radix (PR) is anexterior-resolving herb in traditional Chinese medicine and is also our traditionalfood. The health department announced PR ad "medicine food" in 2000 and in 2002.The old and particular the old women have apt to got osteoporosis than otherpeople. After the women entering menopause, the function of ovary decrease, so thelevel of estrogen decrease quickly and hasten the bone reduction. Estrogenreplacement therapy (ERT) is widely used in preventing post-menopausalosteoporosis. But, this method may increase the risk of developing cancer.Pharmacological studies have revealed that the active ingredients in PR and soybeanare isoflavones, mainly Puerarin of which the aglycone is daidzein. These isoflavoneshave similar structure with estrogen, which are theoretically effective to bonemetabolism. In this study, we observed biochemical markers of bone turnover, bonemineral density (BMD), bone morphometry and changes of element contents in tibiato estimate comprehensively the effect of Isoflavone on bone in ovariectomized rats.The contents of this paper are following:1 Functional studies on the effect of puerarin and daidzein on climacteric osteoporosisFifty Female Sprague Dawley (SD) rats, three -month old, were ovariectomized bilateraliy under anesthesia. Seven days later, animals were orally administrated with daidzein 10mg/kg (6M-OVX-D), puerarin 20mg/kg (6M-OVX-P) or diethylstilbestrol (6M-OVX-E) respectively for 3 months. The 6M-Sham and 6M-OVX group were orally administrated with distilled water. The tibial BMC, femoral BMD, such as the content of calcium, phosphorus, magnesium, znic in tibia and serum, alkaline phosphatase (ALP) activity, were detected after 3 months. Ten Female SD rats, three -month old were detected the above indexes as youthful control group.The result showed that comparied with 6M-OVX group, BMD and calcium content of tibia in 6M-OVX-D and 6M-OVX-E were enhanced respectively (P<0.05).
