Objective: To discuss the pathogenesis of presenile dementia mice .And to discuss the mechanism of action in treating dementia with BDNF.Method: The labyrinth was adopted to test the mice's study memory. The influence during the process of spontaneous activity and consolidation of memory with BDNF micromedicine infusion pump to hippocampus was observed;.The technology of Immunohistochemistry was used to observe the expression of ChaT -IR positive neurons and mean grey were markedly increased in related to encephalic area. The changes of the nucleus basalis of Meynert (NBM) ultrastructure were studied with transmission electron microscope.Results: To test the mice's study memory by Morris water maze latent period (LP) is longer of dementia group than normal's.(~*p<0.01) It was found that LP is longer of without BDNF than treated group's(p<0.05); LP of treated group is different than normal's. The number of CHAT positive neurons increased in encephalic area. And mitochondrial swelling, crest, disappearance obliteration vanish, heterochromatin metachromatin proliferates significantly, arrangement distuebance derangement also appears , neuronal ensemble process swelling, degeneration are manifested as well. BDNF was injected into hippocampi and the pathological changes improve significantly .Conclusion : (1) The degeneration of nucleus basalis is one of the pathological changes of dementialy. (2) The cholinergic neuron's nutriture were changed by BDNF. (3) BDNF significantly enhances ChAT in the related areas. (4) BDNF improves the ultrastructure of cortical neurocyte of...
