Onychomycosis is one of the most frequent nail diseases, its incidence in general population at 2%-15%, which nails are infected by pathogenic fungi. It is chronic infectious and stubborn.Onychomycosis represents up to 50% of nail problems and 30% of all case superfical dermatophytosis. Now, the methods for diagnosing onychomycosis is mainly clinical feasures, direct microscopic examination and fungal culture. However, the positive rate is only 50%-75%. Additonally, the sensitivity of direct microscopy is dependent upon many factors, including the quality and quantity of nail samples, the experience of the operator et al. In fact, it is known there exists about 30% of false-negative results and the sensitivity varies between 25%-80%. It needs at least 4 weeks to identify the strain. All the above restricts the rapid diagnosis and treatment of onychomycosis. More and more new anti-fungal drugs are used to treat onychomycosis, most of the drugs are usually costly and have obvious side-effect. It is necessarily to establish a way for rapid and sensitive diagnosis of onychomycosis .In recent years, with the extentive application of molecular and biologic technique in much field, such as AP-PCR, RFLP, Southern Blotting, PCR is successfully applied in gene study for its good sensitivity, specialties and simpleness.There are a few pathogenic fungi in clinical samples, and it is hard to extract DNA from the samples, people have not made the best use of PCR in clinic.
