OBJECTIVE: To establish Collagen gel droplet culture drug-sensitivity test (CD-DST) technique and explore its clinical feasibility of application.METHODS: The human tumor cell lines were used to find out the best culture conditions of collagen gel droplet culture drug-sensitivity test (CD-DST). The chemosensitivity of 4 or 5 anticancer drugs (ADM, DDP, MMC, PTX, L-OHP, GEM, 5-FU, etc. ) was assessed by the CD-DST in the ovarian and uterine cancer (n=18) , gastrointestinal cancer (n=15) , lymphoma and leukaemia (n=8) , lung cancer (n=5) ,liver cancer (n=4).RESULTS: We established the collagen gel droplet culture drug-sensitivity test (CD-DST) by using the human tumor cell lines. The CD-DST can evaluate the efficacy of anticancer drugs in diverse kinds of tumor specimen, and has good correlation with other chemosensitivity assay methods. Furthermore, the CD-DST has the advantage of using a comparatively small number of cells(3 × 10~3 cell/30μl), which can solve the issue of minor specimen source. There was a statistically correlation between clinically reported response rates and the response rates obtained by CD-DST.
